Directed Evolution of an Artificial Cell Lineage
Biological development is a complex process that mediates between genotypes, to which mutations occur, and phenotypes, on which selection acts. Properties of development can therefore have considerable impact on evolution. However, in many existing simulation models of development, the developmental process itself is difficult to recover and/or analyse. We have previously introduced a model of ...
متن کاملImmobilization of an artificial imine reductase within silica nanoparticles improves its performance.
Silica nanoparticles equipped with an artificial imine reductase display remarkable activity towards cyclic imine- and NAD(+) reduction. The method, based on immobilization and protection of streptavidin on silica nanoparticles, shields the biotinylated metal cofactor against deactivation yielding over 46 000 turnovers in pure samples and 4000 turnovers in crude cellular extracts.
متن کاملDirected evolution of copper nitrite reductase to a chromogenic reductant.
Directed evolution methods were developed for Cu-containing nitrite reductase (NiR) from Alcaligenes faecalis S-6. The PCR cloning strategy allows for the efficient production of libraries of 100 000 clones by a modification of a megaprimer-based whole-plasmid synthesis reaction. The high-throughput screen includes colony lift onto a nylon membrane and subsequent lysis of NiR-expressing colonie...
متن کاملAn (R)‐Imine Reductase Biocatalyst for the Asymmetric Reduction of Cyclic Imines
Although the range of biocatalysts available for the synthesis of enantiomerically pure chiral amines continues to expand, few existing methods provide access to secondary amines. To address this shortcoming, we have over-expressed the gene for an (R)-imine reductase [(R)-IRED] from Streptomyces sp. GF3587 in Escherichia coli to create a recombinant whole-cell biocatalyst for the asymmetric red...
متن کاملDirected evolution of an RNA enzyme.
An in vitro evolution procedure was used to obtain RNA enzymes with a particular catalytic function. A population of 10(13) variants of the Tetrahymena ribozyme, a group I ribozyme that catalyzes sequence-specific cleavage of RNA via a phosphoester transfer mechanism, was generated. This enzyme has a limited ability to cleave DNA under conditions of high temperature or high MgCl2 concentration,...
